The development of new malaria chemoprophylactics is hindered by a significant knowledge gap: liver-stage activity cannot be directly observed in humans. The research team at MMV had developed a sophisticated semimechanistic PKPD model using DSM265 (a DHODH inhibitor) to infer liver-stage growth and initial infection levels by integrating blood-stage and sporozoite-infected human challenge data.
While the modeling was robust, the manuscript faced the hurdle of all high-level pharmacometrics: communicating a multi-stage, multi-compartment model in a way that was both scientifically rigorous for specialists and clear enough for broader clinical stakeholders.

The Solution: Editorial Translation & Strategy
I was brought in to spearhead the editorial development of the manuscript. My work didn’t involve the Monolix simulations, but rather the narrative engineering required to present them.
Structural Refinement: I tidied the initial rough draft, ensuring the transition from the methodology of Induced Blood-Stage Malaria (IBSM) studies to the complexities of the liver-stage model was logically seamless.
Precision in Communication: I refined the presentation of the model’s core equations, such as the turnover model for KillL and KillB, ensuring that parameters like Emax, EC50, and the Hill coefficient (h) were described with absolute clarity.
Peer Review Stewardship: After submission, I managed the revisions following peer review. This involved interpreting reviewer feedback and refining the text to address technical concerns without diluting the paper’s primary message: that liver activity can be inferred to predict clinical outcomes.

The paper was successfully published in CPT: Pharmacometrics & Systems Pharmacology in late 2022. It demonstrated that by assuming similar characteristics between liver and blood-stage activities, researchers could accurately predict the level of chemoprotection afforded by new compounds.

Gene therapy is one of the most complex frontiers in pharmacometrics because it replaces standard mass-balance drug kinetics with biological processes like cellular transduction. Florian Hugi and the team at LYO-X developed a pioneering semimechanistic model to predict human dose-exposure for ADVM-022 (a wAMD gene therapy) by scaling data from non-human primates.
During the initial peer-review phase at Molecular Pharmaceutics, the manuscript faced a critical hurdle. Reviewers were skeptical, with one labeling the research as “premature” due to the inherent novelty of gene therapy scaling. With the LYO-X team facing significant time constraints, the paper required a rapid, high-level strategic intervention to survive the review cycle.

The Solution: Strategic Resubmission & Narrative Defense
I was brought in not just to edit, but to manage the entire submission lifecycle and overhaul the manuscript’s defensive posture.
Addressing Skepticism: I took the lead on the “Response to Reviewers,” carefully reframing the “premature” nature of the data as a vital “first-of-its-kind” framework that the field required for first-in-human dose predictions.
Subscript & Variable Precision: I ensured the rigorous formatting of complex ocular PK variables, such as aqueous clearance (CLaq), vitreous-aqueous elimination (kel,va), and maximal retinal expression (kmax,ret).
- Scaling Narrative: I refined the explanation of how the model decoupled target protein kinetics from the less understood gene therapy scaling, making the animal-to-human translation biologically plausible to the editors.

By taking over the heavy lifting during the revision phase, I helped move the paper from a precarious position to a successful publication in July 2025. While initial pharmacokinetic-pharmacodynamic (PKPD) models provide a foundation for drug activity, they often focus on a single, controlled infection. To better inform late-stage clinical trials, a more robust framework was required, one that could simulate the intensity of natural malaria transmission in endemic regions, accounting for multiple mosquito bites and varying levels of patient treatment adherence.
This project involved reconciling data across a complex ecosystem of stakeholders, including leading global health organizations and multinational pharmaceutical firms, to create a model that could predict Protective Efficacy (PE) in diverse field conditions.

The Solution: Narrative Cohesion for Multi-Stakeholder Research
As the lead editorial consultant, my role focused on the translational bridge between high-level stochastic modeling and regulatory-ready scientific narrative.

Monoclonal antibodies (mAbs) are often engineered with specific mutations (such as LS and YTE) to enhance their affinity for the FcRn receptor, effectively prolonging their circulatory half-life from ~20 days to upwards of 60+ days. While the benefits are clear, predicting exactly how these engineered antibodies will scale from preclinical species (mice and monkeys) to humans remains a significant challenge for drug developers.
A framework was needed that could unify disparate datasets, incorporating wt (wild-type), LS, and YTE variants across multiple species, into a single, predictive model to support early-stage decision-making.

The Solution: From “Blank Page” to Peer-Reviewed Standard
I spearheaded this project from its inception, drafting the manuscript from scratch and managing the technical narrative through the entire development cycle.
- Unified Modeling Narrative: I constructed a narrative that integrated data from human FcRn (hFcRn) Tg32 transgenic mice, cynomolgus monkeys, and clinical human profiles into a single, cohesive population PK model.
Quantifying Mutation Impact: The model successfully quantified that LS and YTE mutations reduce clearance by approximately 69% and 75% respectively , translating to half-life extensions that were statistically conserved across species.
Rigorous Editorial Defense: I managed the submission to a top-tier Taylor & Francis journal, where the work received conditional acceptance with only minor corrections. This validated my ability to present complex allometric scaling and body-weight dependency (coefficients of ~0.74 for clearance) to a highly skeptical peer-review audience.
