The Challenge: A “Premature” Verdict

Gene therapy is one of the most complex frontiers in pharmacometrics because it replaces standard mass-balance drug kinetics with biological processes like cellular transduction. Florian Hugi and the team at LYO-X developed a pioneering semimechanistic model to predict human dose-exposure for ADVM-022 (a wAMD gene therapy) by scaling data from non-human primates.

During the initial peer-review phase at Molecular Pharmaceutics, the manuscript faced a critical hurdle. Reviewers were skeptical, with one labeling the research as “premature” due to the inherent novelty of gene therapy scaling. With the LYO-X team facing significant time constraints, the paper required a rapid, high-level strategic intervention to survive the review cycle.

The Solution: Strategic Resubmission & Narrative Defense

I was brought in not just to edit, but to manage the entire submission lifecycle and overhaul the manuscript’s defensive posture.

  • Addressing Skepticism: I took the lead on the “Response to Reviewers,” carefully reframing the “premature” nature of the data as a vital “first-of-its-kind” framework that the field required for first-in-human dose predictions.

  • Subscript & Variable Precision: I ensured the rigorous formatting of complex ocular PK variables, such as aqueous clearance (CLaq), vitreous-aqueous elimination (kel,va), and maximal retinal expression (kmax,ret).

  • Scaling Narrative: I refined the explanation of how the model decoupled target protein kinetics from the less understood gene therapy scaling, making the animal-to-human translation biologically plausible to the editors.

The Result: Rapid Acceptance & Field Leadership

By taking over the heavy lifting during the revision phase, I helped move the paper from a precarious position to a successful publication in July 2025. The final model successfully predicted human intraocular aflibercept levels observed in the OPTIC Phase 1 trial, proving the utility of the LYO-X framework.

While initial pharmacokinetic-pharmacodynamic (PKPD) models provide a foundation for drug activity, they often focus on a single, controlled infection. To better inform late-stage clinical trials, a more robust framework was required, one that could simulate the intensity of natural malaria transmission in endemic regions, accounting for multiple mosquito bites and varying levels of patient treatment adherence.

This project involved reconciling data across a complex ecosystem of stakeholders, including leading global health organizations and multinational pharmaceutical firms, to create a model that could predict Protective Efficacy (PE) in diverse field conditions.

The Solution: Narrative Cohesion for Multi-Stakeholder Research

As the lead editorial consultant, my role focused on the translational bridge between high-level stochastic modeling and regulatory-ready scientific narrative.

  • Complexity Management: I am responsible for maintaining the narrative thread of the research as it evolves from a liver-stage PK focus to a sophisticated framework incorporating Force of Infection (FOI) and Incidence Rate Ratio (IRR).

  • Stakeholder Stewardship: With lead researchers now at disparate organisations, I act as the central editorial point to ensure the manuscript remains cohesive and maintains the rigorous standards of journals like CPT: Pharmacometrics & Systems Pharmacology.

  • Strategic Sensitivity Analysis: I am currently refining how the manuscript presents sensitivity analyses, particularly regarding assumptions of drug activity during different stages of the parasite lifecycle and the impact of non-adherence on protective outcomes.

The Value: Bridging the Gap to Late-Stage Trials

The output of this project is a modeling framework that allows organisations to swiftly position candidate compounds based on predefined protective efficacy targets. By bringing the modeling closer to a “field perspective,” this work directly informs the design of Phase IIb and Phase III clinical trials.

The Challenge: The Engineering vs. Scaling Gap

Monoclonal antibodies (mAbs) are often engineered with specific mutations (such as LS and YTE) to enhance their affinity for the FcRn receptor, effectively prolonging their circulatory half-life from ~20 days to upwards of 60+ days. While the benefits are clear, predicting exactly how these engineered antibodies will scale from preclinical species (mice and monkeys) to humans remains a significant challenge for drug developers.

A framework was needed that could unify disparate datasets, incorporating wt (wild-type), LS, and YTE variants across multiple species, into a single, predictive model to support early-stage decision-making.

The Solution: From “Blank Page” to Peer-Reviewed Standard

I spearheaded this project from its inception, drafting the manuscript from scratch and managing the technical narrative through the entire development cycle.

  • Unified Modeling Narrative: I constructed a narrative that integrated data from human FcRn (hFcRn) Tg32 transgenic mice, cynomolgus monkeys, and clinical human profiles into a single, cohesive population PK model.
  • Quantifying Mutation Impact: The model successfully quantified that LS and YTE mutations reduce clearance by approximately 69% and 75% respectively , translating to half-life extensions that were statistically conserved across species.

  • Rigorous Editorial Defense: I managed the submission to a top-tier Taylor & Francis journal, where the work received conditional acceptance with only minor corrections. This validated my ability to present complex allometric scaling and body-weight dependency (coefficients of ~0.74 for clearance) to a highly skeptical peer-review audience.

The Result: A “Ready-to-Deploy” Translational Tool

Although the paper is currently undergoing strategic updates before final release, the framework stands as a powerful tool for predicting human PK from preclinical data. It offers developers a way to assess target druggability and dosing feasibility long before the first human dose is ever administered.

In the highly competitive US skincare market, brands must navigate a narrow path between persuasive marketing and the strict regulatory framework of the FDA/FTC cosmetics guidelines. Venture Beyond’s third party required a series of landing pages that leveraged the authority of independent dermatologists to compare premium products against industry giants like Neutrogena and Lancôme.

The challenge was to capture the nuance of expert clinical interviews and “translate” them into a high-converting digital format that adhered to the brand’s specific style guides and U.S. marketing regulations.

The Solution: The “Expert-Witness” Editorial Approach

I served as the editorial bridge between medical expertise and consumer engagement. My process focused on Strategic Transcription & Content Engineering:

  • Clinical Synthesis: I synthesised raw interview data from dermatologists (including Dr. Jessica and Dr. Dana) to ensure that the final copy was a factually accurate reflection of their expert opinions.

A well-established European luxury brand with a 70-year history was struggling to transition from traditional retail to a dominant direct-to-consumer (D2C) presence in the U.S.. While Venture Beyond had achieved remarkable technical results, including a 30% increase in revenue and a 14% growth in contribution margin, there was no cohesive narrative to “sell” this success to stakeholders or future investors.

I was commissioned to interview the growth and creative teams to “extract” the strategy and distill it into a high-impact B2B case study that explained the mechanics of this scaling success.

The Solution: Engineering the “Creative Loop”

I didn’t run the ad accounts, but I was the architect of how this story was told. I translated complex performance metrics into a clear, full-funnel strategy:

  • Pillar 1: Performance-Driven Creative: I articulated how shifting from “branding-heavy” content to “performance-driven” assets (product videos, lifestyle GIFs, and interactive carousels) corrected a critical disconnect in the customer journey.

  • Pillar 2: Growth Strategy & Unit Economics: I detailed the sophisticated financial management that achieved a 35.21% operating profit margin. This involved explaining the move from simple margin-chasing to maximising Total Profit through SKU optimisation and logistics analysis.

  • Pillar 3: AOV & Upselling Mechanics: I documented the three-fold strategy that increased Average Order Value (AOV) by 10%. This included mapping out in-cart and post-purchase upsells that raised the revenue share of high-value luggage sets from 10% to 21%.

The Result: A Historic Milestone

The resulting case study chronicled the brand’s two highest sales dates in history ($267,861 and $215,222 in a single day). By “closing the creative loop” in the narrative, I provided a blueprint for how data-driven media buying can transform a legacy brand into an e-commerce powerhouse.

The Actors’ Trust is a venerable institution with a mission to support professionals in the arts during times of crisis. However, legacy brands often struggle with a “new lease of life”, they need to maintain their historic prestige while appearing modern, approachable, and vital to a younger, digital-native audience.

Working alongside the marketing agency I.E. Digital, I was tasked with writing the copy for the entire website. The objective was to create a cohesive narrative that honoured the Trust’s history while clearly articulating its modern support services to those in need.

To solve a long-standing problem in meta-ethics: how do we use language to express moral “truths” when those truths don’t have a physical, scientific presence in the world? This required bridging the gap between abstract philosophy, neurology, and cognitive psychology to understand how the human brain processes complex, non-literal information.

The Methodology

  • Multidisciplinary Synthesis: Combined linguistic analysis with empirical research from neurology to test how audience psychology dictates the “acceptance” of a narrative.

  • Structural Logic: Developed a framework, Expressive Fictionalism, to explain how language shapes perceptions, emotions, and beliefs.

  • Technical Clarity: Translated dense, theoretical concepts into a 200+ page coherent argument that stood up to rigorous academic defence.
